Methods for cell line generation: | Wild type HEK293T cells were transduced with lentivirus carrying each of the ion channel genes with V5 tag on the C –terminus (as part of the pLX304 vector), then selected with Blasticidin at 10ug/ml, grown in pools and analyzed by anti V5 immunostaining. All cell lines with descriptive info and QC are listed below (cells were marked “pass QC” if V5 was detected or “did not pass” if no V5 was observed). | ||
Cells made by: | Reuben Hogan (cloned lentiviral constructs), UCSF ViraCore (packaged lentivirus), Audre May and Olga Gulyaeva (transduction, culture, cryopreservation, and V5 QC) | ||
Media conditions: | These cells preferably should be grown in standard growth medium (DMEM+ Sodium Pyruvate, Non essential AA, Anti-Anti) supplemented with 10 ug/mL Blasticidin S HCl (Thermo, A1113903) to prevent loss of the integrated construct. | ||
